Compound Inhibition Assay

A compound inhibition assay is an experimental method used to determine whether and how strongly a chemical compound reduces the activity of a biological target, such as an enzyme or receptor. In a typical assay, the target is exposed to increasing compound concentrations under controlled conditions, and changes in a measurable signal reveal inhibition through mechanisms such as competition with a substrate or ligand; dose-response data can yield an IC50 value. These assays support biology and drug discovery by identifying active compounds, comparing potency and selectivity, characterizing molecular interactions, and prioritizing candidates for further validation.

This video demonstrates a viral attachment assay for screening antiviral compounds. Using genetically modified hepatitis C virus and luciferase, it assesses inhibitory effects on viral attachment, as indicated by lower light intensity in co-treated wells compared to wells treated with the virus alone.

This video showcases the Charcoal Agar Resazurin Assay (CARA) for evaluating the activity of antimicrobial compounds against Mycobacterium tuberculosis. CARA aids in assessing the efficacy of these compounds by gauging their impact on bacterial growth and distinguishing between bactericidal and bacteriostatic effects.

An Assay to Measure Influenza Neuraminidase Inhibition Antibody Titers

2025

This video demonstrates an assay measuring influenza neuraminidase (NA) inhibiting antibody titers in a serum sample. A serum sample containing antibodies against NA is added to a fetuin-coated microwell plate. Upon adding the virus, the antibodies prevent viral NA from cleaving fetuin and exposing its galactose moiety. A peroxidase-conjugated lectin is then added, which binds to the exposed galactose. The NA-inhibiting antibody titer is determined using a colorimetric test, where a peroxidase...
