Degenerative Osteoarthritis

Degenerative osteoarthritis is a chronic joint disorder in which cartilage and other joint tissues progressively deteriorate, causing pain, stiffness, and reduced mobility. Mechanical stress, age-related tissue changes, and inflammatory signaling disrupt cartilage homeostasis, while loss of extracellular matrix weakens the joint’s cushioning surface and can promote changes in subchondral bone. Studying this process helps biologists understand interactions among chondrocytes, cartilage, bone, and synovial tissues. This knowledge supports research into risk factors, disease progression, imaging and biomarker development, and treatments aimed at relieving symptoms or preserving joint function.

Osteoarthritis (OA), or degenerative joint disease, is a debilitating condition associated with pain that remains only partially controlled by available analgesics. We describe a protocol for the destabilization of the medial meniscus (DMM) model in mice, an effective tool for osteoarthritis (OA) research. In addition, we have demonstrated that deficiency of progranulincan exaggerate OA development and progression by using this model, indicating that progranulin plays a protective role in the pathogenesis of OA.
