Denaturation Annealing Elongation

Denaturation, annealing, and elongation are the three sequential stages of polymerase chain reaction (PCR), a method used to amplify specific DNA sequences. During denaturation, heat separates double-stranded DNA into single strands; during annealing, the temperature is lowered so short primers bind to complementary target sequences. In elongation, a heat-stable DNA polymerase extends each primer by adding nucleotides to form new complementary strands. Repeating these temperature-controlled cycles produces exponential amplification of the target region, supporting applications in genetic analysis, pathogen detection, cloning, sequencing, and molecular biology research.

Transcription Elongation Factors

2020

Transcription elongation is a dynamic process that alters depending upon the sequence heterogeneity of the DNA being transcribed. Hence, it is not surprising that the elongation complex's composition also varies along the way while transcribing a gene. The transcription elongation is regulated via pausing of RNA polymerase on several occasions during transcription. In bacteria, these halts are necessary because the transcription of DNA into mRNA is coupled to the translation of that mRNA into a...

Gradient Polymerase Chain Reaction to Determine the Optimum Annealing Temperature

2025

This video demonstrates the gradient polymerase chain reaction for optimization of the annealing temperature of primers, which sets gradient annealing temperature along wells. The effect of different annealing temperatures is determined by the yield of amplified products that helps to optimize the reaction condition.

Protein Denaturation

2023

The function of proteins depends on their native three-dimensional structure, which is dictated by the amino acid sequence of the specific protein. Folding of the polypeptide chain takes place under specific conditions that energetically favor the folded conformation. In contrast, protein denaturation occurs spontaneously under unfavorable conditions that disrupt the integrity of the folded conformation. Thus, the chemical and physical environment of a protein, such as significant changes in pH...

Novel Metrics to Characterize Embryonic Elongation of the Nematode Caenorhabditis elegans

2016

Here we detail protocols specifically designed to monitor morphogenic defects that occur during early and late phases of embryonic elongation of the nematode Caenorhabditis elegans. Ultimately, these protocols are designed to identify genes that regulate these phases and to characterize their differential requirements along the antero-posterior axis of the embryo.
