Dura Mater Protection

Dura mater protection refers to the protective role of the dura mater, the tough, fibrous outermost meningeal layer surrounding the brain and spinal cord. Its dense, collagen-rich connective tissue forms a durable barrier, anchors the central nervous system to surrounding structures, and creates cranial folds that help restrict excessive movement; associated dural venous sinuses also support drainage of deoxygenated blood and cerebrospinal fluid. In biology and medicine, understanding this protection clarifies how the meninges reduce mechanical injury, maintain stable neural environments, and influence conditions such as traumatic brain injury, meningitis, dural tears, and intracranial pressure.

Here we present a protocol to visualize spatial correlation of calcitonin gene-related peptide (CGRP)-immunoreactive nerve fibers and blood vessels in the cranial dura mater using immunofluorescence and fluorescent histochemistry with CGRP and phalloidin, respectively. In addition, the origin of these nerve fibers was retrograde traced with a fluorescent neural tracer.
