Exosome Characterization

Exosome characterization is the systematic analysis of small extracellular vesicles commonly called exosomes to determine their physical properties, molecular composition, and biological identity. Researchers assess particle size and concentration using nanoparticle tracking analysis, examine morphology by electron microscopy, and detect characteristic membrane proteins and cargo through methods such as immunoblotting or flow cytometry; exosomes originate from endosomal compartments and are released when multivesicular bodies fuse with the plasma membrane. In biology, these measurements help distinguish exosomes from other extracellular vesicles, evaluate sample quality, and investigate roles in cell-to-cell communication, disease mechanisms, biomarker development, and therapeutic delivery.

This paper demonstrates methods for the isolation, purification and detection of exosomes, as well as techniques for analysis of their molecular content. These methods are adaptable for exosome isolation from both cell culture media and biological fluids, and can beyond analysis of molecular content also be useful in functional studies.

2023

The plasma-isolated exosomes can be used as a non-invasive and repeatable way for the detection and follow-up of the biomarkers for early detection and disease progression. This video describes the protocol for the separation of exosomes from the plasma samples of non-small cell lung cancer patients, which could be used for DNA and protein extractions.
