Fluorochrome Labeling

Fluorochrome labeling is a technique that attaches fluorescent dyes to cells, biomolecules, or molecular probes so they can be detected and localized with light-based instruments. When illuminated at an appropriate excitation wavelength, a fluorochrome absorbs energy and emits light at a longer wavelength; selective chemical coupling or affinity-based binding allows the label to mark targets such as antibodies, nucleic acids, or proteins. In biology, this approach supports fluorescence microscopy, flow cytometry, and immunofluorescence by revealing cellular structures, tracking molecular interactions, and distinguishing cell populations. Its sensitivity and multiplexing potential make it valuable for diagnostics, cell biology, and quantitative research.

This protocol presents a step-by-step guide for the Simultaneous Label-free Autofluorescence Multi-harmonic (SLAM) microscopic technique, including details on how to generate the laser light source, prepare a tissue sample, conduct imaging, and analyze the data. SLAM advances nonlinear microscopy by measuring four complementary label-free contrasts to investigate the tissue microenvironment.

Bromodeoxyuridine Pulse Labelling Assay: A Technique to Measure Cell Proliferation

2023

The video describes the stepwise pulse labeling assay of mammalian cells with bromodeoxyuridine (BrdU) for measuring cell proliferation. The BrdU uptake permits the temporal tracking of cells that were in the synthesis phase at a specific point in time, without requiring the cells to be synchronized.

Metabolic Labeling

2023

Metabolic labeling is used to probe the biochemical transformations and modifications that occur in a cell. This is accomplished by using chemical analogs that mimic the structure of natural biomolecules. Cells utilize analogs in their endogenous biochemical processes, producing compounds that are labeled. The label allows for the incorporation of detection and affinity tags, which can then be used to elucidate metabolic pathways using other biochemical analytical techniques, such as SDS-PAGE...
