Glucose 6-phosphatase

Glucose 6-phosphatase is an enzyme that helps maintain blood glucose by releasing free glucose from glucose 6-phosphate, making it a key regulator of carbohydrate metabolism. Located primarily in the endoplasmic reticulum of liver, kidney, and intestinal cells, it hydrolyzes glucose 6-phosphate to glucose and inorganic phosphate during the final step of gluconeogenesis and glycogenolysis. This reaction allows glucose produced from noncarbohydrate precursors or stored glycogen to enter the bloodstream. Studying glucose 6-phosphatase supports research on metabolic regulation and glycogen storage diseases, including von Gierke disease, which results from impaired enzyme activity.

In this video, we demonstrate an ex vivo technique to measure extracellular glucose analog depletion in harvested mouse organs. It involves incubating the mouse tissue in a solution containing fluorescently-labeled deoxyglucose, which is taken up by the cells. The glucose analog uptake can be indirectly determined by measuring the fluorescence of the incubated medium.
