Granulocyte Colony Stimulating Factor

Granulocyte colony-stimulating factor (G-CSF) is a cytokine that regulates the production, maturation, and release of neutrophils, white blood cells essential for early immune defense. It binds to G-CSF receptors on hematopoietic progenitor cells and mature neutrophils, activating intracellular signaling pathways that promote cell division, differentiation, survival, and movement from bone marrow into the bloodstream. In biology and medicine, G-CSF is used to reduce or treat chemotherapy-associated neutropenia and to mobilize hematopoietic stem cells for collection before transplantation. Studying G-CSF also clarifies how soluble signals control blood-cell formation and coordinate innate immune responses.

Here, we present a protocol to differentiate murine granulocyte-macrophage-colony-stimulating-factor-producing T helper (THGM) cells from naive CD4+ T cells, including isolation of naive CD4+ T cells, differentiation of THGM, and analysis of differentiated THGM cells. This method can be applied to studies of the regulation and function of THGM cells.

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.
