Heterochromatin Protein 1

Heterochromatin Protein 1 (HP1) is a family of chromatin-associated proteins that helps organize and maintain compact, transcriptionally silent regions of the genome. Its chromodomain recognizes methylated lysine 9 on histone H3 (H3K9me), while its chromoshadow domain promotes dimerization and recruits additional chromatin-modifying or structural factors, enabling heterochromatin assembly and propagation. In biology, HP1 is used to investigate gene regulation, chromosome stability, DNA repair, and epigenetic inheritance, with studies examining how altered HP1 interactions influence development, genome integrity, and disease-related changes in chromatin organization.

The extent of chromatin compaction can be studied by staining chromatin using specific DNA binding dyes. Under the microscope, the dense-compacted regions that take up more dye are called heterochromatin. Heterochromatin is further classified into two forms – constitutive heterochromatin and facultative heterochromatin. Constitutive heterochromatin: It is a highly compact region of chromatin that is mostly concentrated in the centromere and telomere. Unlike euchromatin, the amino acid at 9th...

Here, we describe some established methods to determine endoplasmic reticulum (ER) stress and unfolded protein response (UPR) activation, with particular emphasis on HIV-1 infection. This article also describes a set of protocols to investigate the effect of ER stress/UPR on HIV-1 replication and virion infectivity.
