High-density Colony Arrays

High-density colony arrays are organized collections of microbial colonies grown in closely spaced, addressable positions on a solid surface, enabling many biological samples to be analyzed in parallel. Cells are deposited or transferred into defined locations, where each colony expands from a small population under controlled growth conditions while remaining spatially separated for identification, screening, or recovery. In biology, these arrays support high-throughput genetic screening, strain comparison, library characterization, and selection of organisms with desired traits. Their ordered format improves experimental efficiency, reduces reagent use, and links observable colony phenotypes with specific samples or genetic variants.

(2021)This video demonstrates the use of photodegradable hydrogels to isolate bacterial colonies within silicon microwell arrays. Bacterial cells are first loaded into microwells, encapsulated in a crosslinked hydrogel, and cultured into distinct colonies. Upon exposure to ultraviolet light, targeted degradation of the hydrogel enables selective extraction of individual...

We report a simple method for fabricating an ultrahigh density array of vertically ordered small-molecular organic nanowires. This method allows for synthesis of complex heterostructured hybrid nanowire geometries, which can be inexpensively grown on arbitrary substrates. These structures have potential applications in organic electronics, optoelectronics, chemical sensing, photovoltaics and spintronics.

Keratinocyte Colony Formation Assay: An In Vitro Assay to Assess the Ability of Keratinocytes to Grow into Colonies

2023

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.
