Hypoxia Inducible Factors

Hypoxia-inducible factors (HIFs) are oxygen-sensitive transcription factors that help cells adapt to limited oxygen, a condition known as hypoxia. Under normal oxygen levels, prolyl hydroxylation marks HIF-alpha subunits for recognition by the von Hippel-Lindau protein and subsequent degradation; during hypoxia, this modification decreases, allowing HIF-alpha to stabilize, dimerize with HIF-beta, and bind hypoxia-response elements in DNA. HIF signaling regulates genes involved in glycolysis, angiogenesis, erythropoiesis, and cellular survival. Studying these factors helps explain physiological adaptation and disease processes, including cancer, ischemia, and anemia, while informing research into therapies that modulate oxygen-sensing pathways.
