Ice Cold Pbs

Ice-cold PBS, or phosphate-buffered saline, is a chilled, isotonic solution used to handle and wash biological samples while preserving cellular and molecular structure. Its phosphate buffer helps maintain a near-physiological pH, while sodium chloride and related salts support osmotic balance; cooling the solution reduces enzymatic activity and slows unwanted biochemical changes. In biology laboratories, ice-cold PBS is commonly used to rinse cultured cells, remove media or residual reagents, and maintain samples during protein extraction, immunostaining, and cell-based assays. Proper use can improve sample integrity, reduce degradation, and promote consistent results across downstream experiments.

Here, we present a protocol to demonstrate the generation of ice when water is introduced to a cold bath of brine, as a secondary refrigerant, at a range of temperatures well below the freezing point of water. It can be used as an alternative way of producing ice for industry.

Antifreeze proteins (AFPs) bind to specific planes of ice to prevent or slow ice growth. Fluorescence-based ice plane affinity (FIPA) analysis is a modification of the original ice-etching method for determination of AFP-bound ice planes. AFPs are fluorescently labeled, incorporated into macroscopic single ice crystals, and visualized under UV light.
