Islet Live Imaging

Islet live imaging is the real-time visualization of living pancreatic islets, the endocrine cell clusters that regulate blood glucose, and it provides a direct way to study coordinated cell behavior. Using time-lapse microscopy with fluorescent indicators or genetically encoded reporters, researchers monitor changes such as intracellular calcium dynamics, hormone release, and interactions among insulin-producing beta cells under controlled conditions. This approach reveals how islets respond to glucose and other stimuli, helping clarify mechanisms of insulin secretion, beta-cell communication, and islet dysfunction. Its applications include diabetes research, assessment of therapeutic compounds, and evaluation of engineered or transplanted islet tissue.

This video describes the technique of imaging islet cells in real-time, to study the changes in cytosolic calcium concentration in the minor subpopulations of the pancreatic islet cells. Minor cell subpopulations can be identified using functional responses to marker compounds, such as adrenaline and ghrelin, that induce changes in cytosolic calcium in a specific populations of islet cells. This can provide an insight into the cross talk between the blood glucose and the islet secretory output.

We describe a protocol for visualization of insulin exocytosis in intact islets using pHluorin, a pH-sensitive green fluorescent protein. Isolated islets are infected with adenovirus encoding pHluorin coupled to the vesicle cargo neuropeptide Y. This allows for the detection of insulin granule fusion events by confocal microscopy.

Pancreatic islets, also called the Islets of Langerhans, are a cluster of endocrine cells which produces hormones for glucose regulation and other important biological functions. This protocol explains a method for isolating pancreatic islets by first perfusing collagenase via Ampulla of Vater to digest the exocrine pancreatic tissue and a simplified density gradient centrifugation to purify the islets from mice.
