Lens Suture Analysis

Lens suture analysis is the microscopic or imaging-based examination of the seams formed where lens fibers meet, providing a way to study the organization and development of the vertebrate eye lens. As new fiber cells differentiate from the lens epithelium, they elongate, lose their nuclei and organelles, and align so their ends converge into characteristic suture patterns; analyzing these patterns can reveal changes in lens structure. In biology, this approach supports research on lens morphogenesis, maturation, aging, and disorders such as cataract, linking cellular architecture with optical function and disease-related disruption.

Lens development involves interactions with other tissues. Several zebrafish eye mutants are characterized by an abnormally small lens size. Here we demonstrate a lens transplantation experiment to determine whether this phenotype is due to intrinsic causes or defective interactions with tissues that surround the lens.

We present an efficient method of studying lens accommodation by using a manual lens stretcher. The protocol mimics physiological accommodation by pulling the zonules connected around the lens capsule, thereby, stretching the lens.
