Luminometer

A luminometer is an instrument that measures the intensity of light produced by luminescent chemical or biological reactions, providing sensitive quantitative data in biology. In a typical assay, a sample generates bioluminescence or chemiluminescence, and a photodetector converts emitted photons into an electrical signal that the instrument records as relative or absolute light output. Luminometers support ATP detection, enzyme activity measurements, cell viability assays, and reporter-gene analysis, including luciferase-based studies of gene expression. By linking light intensity to reaction or cellular activity, they enable rapid, noninvasive measurements and help researchers evaluate metabolism, signaling, toxicity, and other biological processes.

We describe an alternative approach to the enumeration of mycobacteria in vitro, which uses reporter-gene tagged mycobacteria instead of colony-forming units (CFU). “Survival” of organisms as well as host response-markers are measured simultaneously, providing a low-cost, versatile and functional system for studies of host/pathogen interactions in the context of tuberculosis.
