Male Sterile Mutants

Male sterile mutants are organisms with genetic changes that prevent the production or release of functional male gametes, while female reproductive function may remain intact. In plants, mutations can disrupt anther formation, meiosis, pollen development, or pollen maturation, producing flowers that cannot fertilize themselves but can receive pollen from another plant. These mutants are valuable in biology for studying reproductive development, gene function, and fertility regulation. In crop breeding, male sterility supports controlled cross-pollination and hybrid seed production by reducing the need for manual removal of anthers, helping researchers and breeders generate predictable combinations of parental traits.

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN In 1959 The 3 R's were introduced by W.M.S. Russell and R.L. Burch in their book The Principles of Humane Experimental Technique. The 3 R's are replacement, reduction, and refinement of the use of animals in research. The use of cell lines and tissue cultures that originated from research animals is a replacement technique, as it allows for many experiments to be conducted in vitro. Harvesting...

Enriching for Males: Maintaining a Stock of Male C. elegans

Nematodes can be propagated easily in the lab--in part because hermaphrodites can reproduce by self-fertilization. However, genetic crosses and other experiments may require a large number of males. This video describes a method to enrich for male C. elegans.

The sterile insect technique (SIT) is used to control specific, medically important mosquito populations that may be resistant to chemical controls. Here, we describe a method of mass rearing and preparation of sterile male mosquitoes for release in an operational SIT program targeting the Aedes aegypti mosquito.
