Mosaic Transgenesis

Mosaic transgenesis is a genetic engineering approach that produces an organism composed of cells with different genotypes, including transgene-bearing and unmodified cells. It typically occurs when a transgene is introduced into a fertilized egg or early embryo after some cell divisions have begun, so only the targeted cell and its descendants inherit the construct. The resulting patchwork of gene expression can reveal when and where a gene functions during development, while fluorescent reporters enable visualization of affected lineages. In biology, mosaic transgenesis supports cell-fate mapping, tissue-specific gene analysis, disease modeling, and studies of genetic function when whole-organism modification is impractical or disruptive.

Transgenic manipulations and genome editing are critical for functionally testing the roles of genes and cis-regulatory elements. Here a detailed microinjection protocol for the generation of genomic modifications (including Tol2-mediated fluorescent reporter transgene constructs, TALENs, and CRISPRs) is presented for the emergent model fish, the threespine stickleback.

In model organisms, transgenesis can manipulate gene functions while RNAi can knockdown specific mRNA transcripts 1-2. This protocol aims to illustrate the techniques needed to introduce stably transmitted DNA and transient double stranded RNA into the necromenic nematode Pristionchus pacificus for studies in evolutionary, developmental, and behavioral biology.

Fluid Mosaic Model

2026

The fluid mosaic model was first proposed as a visual representation of research observations. The model comprises the composition and dynamics of membranes and serves as a foundation for future membrane-related studies. The model depicts the structure of the plasma membrane with a variety of components, which include phospholipids, proteins, and carbohydrates. These integral molecules are loosely bound, defining the cell’s border and providing fluidity for optimal function.LipidsThe most...
