Necrosis Induction

Necrosis induction is the deliberate triggering of necrotic cell death, a form of irreversible cellular injury that disrupts tissue integrity and can provoke inflammation. It typically results from severe stress such as toxin exposure, physical damage, infection, or loss of oxygen and energy, causing ATP depletion, failure of ion regulation, cellular swelling, plasma-membrane rupture, and release of intracellular contents. In biology, necrosis induction helps researchers examine cell-death pathways, inflammatory signaling, tissue injury, and host responses to environmental or microbial stress. Controlled experimental models also support studies of disease mechanisms and evaluation of compounds that protect cells or promote targeted cell destruction.

Necrosis is considered as an “accidental” or unexpected form of cell death that ends in cell lysis. The first noticeable mention of “necrosis” was in 1859 when Rudolf Virchow used this term to describe advanced tissue breakdown in his compilation titled “Cell Pathology”. Morphological Manifestations of Necrosis Necrotic cells show different types of morphological appearance depending on the type of tissue and infection. In coagulative necrosis, cells become anucleated and die, but their...

Source: Chen, Y. et. al., Evaluating Virulence and Pathogenesis of Aeromonas Infection in a Caenorhabditis elegans Model. J. Vis. Exp. (2018)This video demonstrates a muscle necrosis assay in C. elegans (Caenorhabditis elegans) using pathogenic Aeromonas strains. By feeding green fluorescent protein (GFP)-expressing worms on Aeromonas-seeded plates, the assay models bacterial infection and reveals tissue damage through fluorescence imaging. The progressive loss of muscle structure and GFP...

Necrosis is a form of irreversible cell death caused by severe injury such as ischemia, toxins, or trauma. Unlike programmed cell death, it is an uncontrolled, pathological process that typically provokes inflammation in surrounding tissues.Pathophysiologic ChangesNecrosis begins when cells sustain critical damage, leading to swelling of organelles, particularly mitochondria, and rapid ATP depletion. As energy levels decline, membrane ion pumps fail, leading to calcium influx and eventually,...

Source: Yong P. Chen, PhD, Department of Physics & Astronomy, College of Science, Purdue University, West Lafayette, IN This experiment will use inductive coils to demonstrate the concept of inductor and inductance. Magnetic induction will be demonstrated using a rod magnet inserted into or extracted away from the core of a coil to induce a transient electromotive force (emf) voltage in the coil, measured by a voltmeter. This experiment will also demonstrate the mutual inductance between...

University of Notre Dame, IN The Guide for the Care and Use of Laboratory Animals ("The Guide") states that pain assessment and alleviation are integral components of the veterinary care of laboratory animals. The definition of anesthesia is the loss of feeling or sensation. It is a dynamic event involving changes in anesthetic depth with respect to an animal's metabolism, surgical stimulation, or variations in the...
