Op50 E Coli Feeding

OP50 E. coli feeding is a laboratory method for maintaining and studying the nematode Caenorhabditis elegans, using bacteria as a standardized food source. Researchers grow the uracil-requiring OP50 strain on agar plates, where worms ingest the bacterial lawn through pharyngeal pumping; its limited growth helps provide a relatively thin, manageable food layer for observing development and behavior. This approach supports routine C. elegans culture, genetic experiments, and investigations of reproduction, metabolism, aging, and host-microbe interactions. Because feeding conditions can influence worm physiology, consistent OP50 preparation is important for reproducible biological studies.

Behavioral assays provide powerful tools for understanding neuronal function. Here we present several protocols for quantifying predatory feeding behavior found in the model nematode Pristionchus pacificus and its relatives. Additionally, we provide methods for analyzing predatory feeding adaptations including mouth structures and teeth.
