Pcr Product Ligation

PCR product ligation is a molecular cloning technique that joins an amplified DNA fragment to a compatible vector, enabling the fragment to be propagated or analyzed in biological research. DNA ligase catalyzes formation of phosphodiester bonds between adjacent 5′ phosphate and 3′ hydroxyl groups after complementary cohesive ends or compatible overhangs align; some workflows use added restriction sites or single-base overhangs for directional or TA cloning. The resulting recombinant plasmid can be introduced into host cells for sequence verification, gene expression, mutation analysis, or construction of larger DNA assemblies.
