Peg-mediated Transfection

PEG-mediated transfection is a method for introducing exogenous nucleic acids, such as plasmid DNA, into cells by using polyethylene glycol (PEG) to promote cellular uptake. PEG changes the physical interactions between nucleic acids and the cell surface and can temporarily perturb or promote fusion of cellular membranes, allowing genetic material to cross the membrane and reach the cytoplasm; treatment conditions must then be controlled to limit cell damage. In biology, the method supports transient gene expression, genetic manipulation of otherwise difficult-to-transfect cells, and studies of gene function, while its efficiency depends on PEG concentration, exposure time, cell type, and recovery conditions.
