Plant Metabolomics

Plant metabolomics is the comprehensive study of small molecules, or metabolites, produced by plants and how their levels change across tissues, developmental stages, and environmental conditions. Metabolites are extracted from plant samples and profiled using analytical platforms such as liquid or gas chromatography coupled with mass spectrometry, or nuclear magnetic resonance spectroscopy; statistical and bioinformatic analysis then links metabolite patterns to biological processes. Plant metabolomics helps reveal metabolic pathways, characterize responses to drought, pathogens, or nutrients, and identify biomarkers associated with plant traits. These data support crop improvement, food quality assessment, and studies of plant adaptation and physiology.

This paper presents the use of PSYCHE and SAPPHIRE-PSYCHE in the metabolic profiling of plants and includes detailed procedures for sample preparation and optimal Pure Shift NMR spectra recording. Examples through which the gain in resolution achieved by homonuclear decoupling allows a more comprehensive understanding of the system are discussed.

Metabolite profiling has been a valuable asset in the study of metabolism in health and disease. Utilizing normal-phased liquid chromatography coupled to high-resolution mass spectrometry with polarity switching and a rapid duty cycle, we describe a protocol to analyze the polar metabolic composition of biological material with high sensitivity, accuracy, and resolution.
