Primary Fibroblast Culture

Primary fibroblast culture is the laboratory maintenance and expansion of fibroblasts isolated directly from tissue, providing a model for studying connective-tissue biology and cellular responses. Tissue samples are mechanically and often enzymatically dissociated, after which fibroblasts attach to a culture surface and proliferate in nutrient-supplemented medium under controlled temperature, pH, and gas conditions; researchers must monitor confluence, contamination, and passage number to preserve experimental consistency. These cultures support investigations of extracellular matrix production, wound healing, fibrosis, cell signaling, and tissue repair, while retaining donor- and tissue-specific features that can complement immortalized cell lines in disease modeling, drug testing, and regenerative biology.

We describe a simple and quick experimental procedure for generating primary fibroblasts from the ears and tails of mice. The procedure does not require special animal training and can be used for the generation of fibroblast cultures from ears stored at RT for up to 10 days.

Naïve Adult Stem Cells Isolation from Primary Human Fibroblast Cultures

We report a method to isolate naïve multipotent skin-derived precursor (SKP) cells from primary human fibroblast cultures. We show that these SKPs derived from fibroblast cultures share similar stem cell properties to the ones derived directly from human skin biopsies. These cells express the neural crest marker, nestin, in addition to the multipotent markers such as OCT4 and Nanog.
