Ratio-metric Dye

A ratio-metric dye is a fluorescent or colorimetric indicator that reports a biological or chemical condition by comparing signals measured at two wavelengths rather than relying on a single intensity. Binding to a target analyte, changes in pH, or other environmental shifts alter the dye’s spectral properties, changing the ratio between its emission or absorbance signals. Because this measurement helps compensate for dye concentration, illumination, and optical-path differences, ratio-metric dyes can provide more reliable quantitative data in biological samples. Researchers use them to monitor intracellular calcium, pH, ion concentrations, and related cellular processes in living cells and tissues.
