Septin Mislocalization

Septin mislocalization is the abnormal redistribution of septins, GTP-binding cytoskeletal proteins that organize cell membranes and support processes such as cytokinesis and cell polarity. Septins normally assemble into filaments and higher-order structures at specific cellular sites through regulated interactions with membranes, proteins, and other cytoskeletal components; changes in these targeting cues can place them in inappropriate compartments. This disruption can impair cell shape, division, migration, and intracellular organization. Studying septin mislocalization helps explain how altered cytoskeletal architecture contributes to developmental abnormalities, cancer, neurodegeneration, and pathogen infection, while providing potential markers of cellular dysfunction and targets for therapeutic research.

Septins

Septins are protein filaments forming the cytoskeleton along with the microtubules, microfilaments, intermediate filaments, and other accessory proteins. In 1971 while studying the cell division cycle in mutant Saccharomyces cerevisiae Harwell et al. first identified the septin-related genes playing a crucial role in yeast cytokinesis. Fluorescence microscopy revealed that these proteins localize at the budding neck as rings. These ring-like proteins were then named Septins by John Pringle, and...

2025

This video demonstrates an in vitro protocol to study the Msp1-mediated removal of mislocalized TA proteins integrated into the lipid bilayer of liposomes. Msp1 and TA proteins are co-reconstituted into liposomes, and the ATP-dependent translocation of TA proteins is confirmed through affinity purification.

Role of Septins

Septins are the recently discovered fourth major protein component of the cytoskeleton, along with microfilaments, microtubules, and intermediate filaments. These proteins can associate with other cytoskeletal filaments and carry out varied roles or can be free-floating in the cytoplasm. Cellular Functions of Septins Recent studies have revealed the multifaceted roles of septins in various cellular processes such as cytokinesis, ciliogenesis, and neurogenesis. Septins act as scaffolds and...
