Sterile Ecm Preparation

Sterile ECM preparation is the controlled processing of extracellular matrix (ECM) materials to remove contaminants while preserving the biological structures and signals that support cell behavior. It typically combines aseptic handling with validated sterilization or decontamination steps, followed by preparation of the matrix as a coating, hydrogel, or scaffold under conditions that limit microbial growth and unwanted degradation. Properly prepared ECM provides cells with attachment sites and biochemical cues, making it valuable for cell culture, tissue engineering, regenerative biology, and studies of cell adhesion, migration, differentiation, and tissue organization.

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN In 1959 The 3 R's were introduced by W.M.S. Russell and R.L. Burch in their book The Principles of Humane Experimental Technique. The 3 R's are replacement, reduction, and refinement of the use of animals in research. The use of cell lines and tissue cultures that originated from research animals is a replacement technique, as it allows for many experiments to be conducted in vitro. Harvesting...

PTM Based Tumor Microenvironment Monitoring: A Method for Tracking Changes in ECM Rigidity Using Tumor Spheroids

This protocol describes a technique for monitoring extracellular matrix rigidity changes by using particle-tracking microrheology (PTM). This is done by culturing 3D pancreatic tumor spheroids into ECM with embedded fluorescent probes for video particle-tracking and analysis.

Cleaning, Sterilization, and Disinfection

Cleaning, disinfection, and sterilization are the methods that help to break the infection chain and prevent disease. Cleaning The cleaning process usually involves using water with detergents or enzymatic cleaner and removing foreign material from objects and surfaces, including organic material such as body fluids or inorganic material like soil. Cleaning is performed before high-level disinfection and sterilization because foreign materials on the cover of the devices interfere with process...
