Tissue Culture Insert

A tissue culture insert is a laboratory device that supports cells on a porous membrane suspended within a culture well, allowing researchers to model interactions between distinct cellular compartments. The membrane separates apical and basolateral media while permitting diffusion of nutrients, signals, and experimental compounds; cells can form polarized layers or biological barriers at the interface. In biology, tissue culture inserts are used to study epithelial permeability, transport, migration, co-culture interactions, and tissue responses to drugs or pathogens. By enabling controlled sampling from each compartment, they improve analysis of cell communication and barrier function in models such as intestinal, respiratory, and vascular tissues.

Whole Organ Tissue Culture

Whole organs can be cultured ex vivo using specialized bioreactors with the goal of repairing or replacing entire organs. This method uses a donor organ that is stripped of all cells, leaving behind the three-dimensional structure, which is then repopulated with new cells. This video demonstrates the whole organ culture of lungs and shows how a dynamic culture that mimics the mechanical stimulation in the body is needed to induce native tissue properties.

Although two-dimensional tissue culture has been common for some time, cells behave more realistically in a three-dimensional culture, and more closely mimics native tissue. This video introduces histotypic tissue culture, where the growth and propagation of one cell line is done in an engineered three-dimensional matrix to reach high cell density. Here, we show the harvesting of cells from donor tissue, followed by cell culture on an engineered construct.

Tissue culture inserts with plastic membranes are the golden standard in cell culture labs as permeable supports to establish cell layers and models of barrier tissues. Herein, we present a simple method to replace the plastic membrane with a more biologically relevant membrane made from a recombinant functionalized spider silk protein.
