Tissue Microdissection

Tissue microdissection is a technique for isolating defined cells or microscopic regions from a tissue section, enabling molecular analysis while retaining anatomical context. Under microscopic guidance, researchers identify target areas and separate them from surrounding tissue using a fine blade, needle, or focused laser, then collect the material for downstream extraction of DNA, RNA, or proteins. In biology, this approach helps distinguish molecular differences among neighboring cell populations within complex tissues. Applications include gene-expression studies, genomic and proteomic analyses, developmental research, and investigation of disease-associated changes that may be obscured in bulk tissue samples.

In this video-article we present a method for isolating single or multiple Drosophila da neurons from third instar larvae using the infrared capture (IR) class of Laser Capture Microdissection (LCM). RNA obtained from the isolated neurons can be readily used for downstream applications including qRT-PCR or microarray analyses.
