Two-photon Laser

A two-photon laser is a pulsed light source used to drive two-photon excitation, a process that enables fluorescence imaging with longer-wavelength light and is especially valuable in biological research. At the focal point of a high-intensity, typically femtosecond near-infrared beam, a fluorescent molecule simultaneously absorbs two lower-energy photons and then emits a shorter-wavelength photon. Because this nonlinear absorption occurs mainly at the focal plane, two-photon microscopy produces optical sectioning with limited out-of-focus excitation and reduced photodamage. The technique supports high-resolution imaging of living cells, tissues, and neuronal activity, including studies in relatively deep and scattering biological samples.
