Water Sample Isolation

Water sample isolation is the process of separating microorganisms or other biological components from water so they can be detected, characterized, or studied under controlled laboratory conditions. In biology, researchers collect samples aseptically and concentrate their contents through methods such as membrane filtration, centrifugation, or serial dilution, followed by cultivation on selective or general-purpose media when viable organisms are the target. Isolated colonies or recovered cells can then support microbial identification, environmental monitoring, water-quality assessment, and research on community composition, contamination, and ecological function. Careful sampling and contamination control are essential for producing results that accurately represent the original water source.

Source: Laboratories of Dr. Ian Pepper and Dr. Charles Gerba - The University of Arizona Demonstrating Author: Luisa Ikner The quality of water destined for use in agricultural, recreational, and domestic settings is of great importance due to the potential for outbreaks of waterborne disease. Microbial agents implicated in such events include parasites, bacteria, and viruses that are shed in high numbers in the feces of infected people and animals. Transmission to new and susceptible hosts may...

Source: Cross, T., et al. An Optimized Enrichment Technique for the Isolation of Arthrobacter Bacteriophage Species from Soil Sample Isolates. J. Vis. Exp.(2015)This video demonstrates the isolation of bacteriophages from a soil sample. A soil sample is collected and suspended in a phage buffer and allowed to settle the sediment. The extract is filtered, and nutrients are added. A vacuum filtration is done to remove microbial contaminants. The filtrate with phages is aliquoted into flasks, and...
