Mesothelial Cells

Mesothelial cells are specialized epithelial cells that form the mesothelium, a protective lining around body cavities and organs such as the pleura, peritoneum, and pericardium. They create a selectively permeable barrier and secrete lubricating substances that reduce friction, while injury or inflammatory signals can stimulate repair, phenotypic changes, and epithelial-to-mesenchymal transition. In cancer research, mesothelial cells are studied as potential origins of mesothelioma and as active participants in tumor microenvironments. Their interactions with circulating tumor cells can influence adhesion, invasion, metastasis, and treatment response, making them relevant targets for understanding serosal cancers and cancer dissemination.

Ovarian cancer cell invasion into the mesothelial lining of the peritoneum is a dynamic process over time. Utilizing a real time analyzer, the invasive capacity of ovarian cancer cells in a spheroid-mesothelial cell co-culture model can be quantified over prolonged time periods, providing insights into factors regulating the metastatic process.

Proximal Culture System: An In Vitro Culture Technique to Study Paracrine Signaling Between Cells

In this video, we develop a proximal co-culture model of ovarian cancer cells and primary human mesothelial cells. This physiologically relevant model allows the study of paracrine signaling, where the localized concentrations of the secreted factors are maintained while preventing direct cellular contact.
