Palbociclib

Palbociclib is a selective cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor that slows cancer cell proliferation by disrupting cell-cycle progression. It blocks CDK4/6-mediated phosphorylation of the retinoblastoma protein, maintaining this growth-regulating protein in an active state and preventing tumor cells from advancing from the G1 phase into S phase. In cancer research, palbociclib is used primarily with endocrine therapy to treat hormone receptor-positive, HER2-negative advanced or metastatic breast cancer. Its effects also support studies of cell-cycle control, treatment resistance, combination therapies, and biomarkers that may predict response or toxicity.
