Alkaline Detergent

Alkaline detergent is a cleaning formulation with a basic pH that removes organic residues, oils, and particulate soil from surfaces. Its alkalinity promotes the hydrolysis and loosening of fats and proteins, while surfactants lower surface tension and emulsify or suspend released contaminants in water. Builders may bind metal ions and improve cleaning performance, especially in hard water. In chemistry and laboratory practice, alkaline detergents are used to clean glassware, instruments, industrial equipment, and processing surfaces, where their effectiveness must be balanced against potential corrosion or damage to alkali-sensitive materials.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.

Enrichment of Bacterial Lipoproteins Using Non-Ionic Detergent Phase Separation

(2018)This video demonstrates the enrichment of bacterial lipoproteins using a non-ionic detergent that undergoes phase separation upon warming. Through sequential temperature shifts and centrifugation steps, lipoproteins are selectively partitioned into the detergent phase, while non-lipoprotein contaminants...

Alkaline Single Cell Gel Electrophoresis: A Sensitive Technique for Quantitative Estimation of DNA Damage in Individual Cancer Cells Following Chemotherapy

2025

In this video, we demonstrate an alkaline single-cell electrophoresis assay to quantify both single- and double-strand DNA breaks in doxorubicin-treated cells. Alkaline conditions disrupt the hydrogen bonds between DNA base pairs, resulting in the complete unwinding and separation of DNA strands. This allows differential migration of undamaged and damaged DNA fragments in agarose gel upon electrophoresis, revealing a comet-shaped structure of slowly migrating undamaged DNA forming the circular...

Detergent Purification of Membrane Proteins

2023

Detergents are used to purify the integral proteins of the membrane. The hydrophobic portion of the detergent can replace membrane phospholipids while solubilizing the membrane proteins. When detergent monomers reach a specific concentration in a solution called critical micelle concentration (CMC), they form micelles. Above CMC, the concentration of the detergent monomers remains in equilibrium with the micelle. The number of detergent monomers present in the CMC varies for each detergent, and...
