Density Refractive Index Matching

Density refractive index matching is a materials preparation method that adjusts a liquid medium to match both the density and refractive index of suspended particles or another phase. By selecting solutes and their concentrations, researchers can reduce density differences that drive sedimentation while minimizing light bending and scattering at interfaces. This combined control makes complex systems more accessible to optical observation and quantitative analysis. In chemistry, the method supports investigations of colloidal dispersions, particle transport, sedimentation, mixing, and interfacial behavior, improving measurements of structure and dynamics in complex fluids.

This protocol describes the fabrication of microfluidic devices from MY133-V2000 to eliminate artifacts that often arise in microchannels due to the mismatching refractive indices between microchannel structures and an aqueous solution. This protocol uses an acrylic holder to compress the encapsulated device, improving adhesion both chemically and mechanically.

Reflection and Refraction

Source: Derek Wilson, Asantha Cooray, PhD, Department of Physics & Astronomy, School of Physical Sciences, University of California, Irvine, CA Light travels at different speeds depending on the material through which it is propagating. When light travels from one material to another, it will either slow down or speed up. In order to conserve energy and momentum, the light must change the direction in which it propagates. This bending of light is known as refraction. Some fraction of the...

Sign Test for Matched Pairs

2025

The sign test for matched pairs offers a robust method for comparing two paired samples, often for the effects of an intervention in one of them. This method is very useful in situations where the underlying distribution of the data is unknown. The test compares two related samples—often pre- and post-treatment measurements on the same subjects—to determine if there are significant differences in their median values. To conduct the sign test, we first calculate the differences in value between...

Determining the Density of a Solid and Liquid

Source: Laboratory of Dr. Michael Evans — Georgia Institute of Technology The ratio of the mass of a substance to its volume is known as the mass density or, simply, the density of the substance. Density is expressed in units of mass per volume, such as g/mL or kg/m3. Because the density of a substance does not depend on the amount of substance present, density is an “intensive property”. To measure the density of a sample of material, both the mass and volume of the sample must be determined.

Density Gradient Ultracentrifugation

2023

Density gradient ultracentrifugation is a common technique used to isolate and purify biomolecules and cell structures. This technique exploits the fact that, in suspension, particles that are more dense than the solvent will sediment, while those that are less dense will float. A high-speed ultracentrifuge is used to accelerate this process in order to separate biomolecules within a density gradient, which can be established by layering liquids of decreasing density in a centrifuge tube. The...
