Pathlength Correction

Pathlength correction is the adjustment of an optical measurement to account for the distance light travels through a sample, allowing results collected with different sample geometries to be compared. In spectrophotometry, the correction uses the Beer–Lambert relationship, in which absorbance depends on pathlength, analyte concentration, and molar absorptivity; measured absorbance can therefore be rescaled to a defined reference pathlength. This approach improves quantitative analysis when environmental samples are measured in microvolume devices, nonstandard cuvettes, or variable-depth containers. Accurate correction supports reliable estimates of nutrients, pollutants, and other dissolved substances in water and related environmental matrices.
