Threshold Cycle Measurement

Threshold cycle measurement is a quantitative real-time PCR method that estimates the starting amount of a target nucleic acid by recording the amplification cycle at which fluorescence exceeds a defined threshold. During PCR, target DNA is repeatedly denatured, annealed, and extended, while fluorescent dyes or probes generate a signal that rises with product accumulation; lower threshold cycle values generally indicate more starting template. In environmental sciences, this measurement supports detection and quantification of microbial genes, pathogens, and indicators in water, soil, and other samples. It helps assess contamination, monitor ecosystem changes, and compare molecular targets across environmental conditions.

Here, we describe a rapid reliable and simple procedure to determine the lowest temperature at which rats or mice show nocifensive behavior, i.e. the thermal nociceptive threshold (TNT).
