Guide Rna Design

Guide RNA design is the process of selecting and engineering an RNA sequence that directs a CRISPR-associated nuclease to a chosen genetic locus, a key step in targeted genome editing. The guide region base-pairs with complementary DNA near a compatible protospacer-adjacent motif, positioning the nuclease for sequence-specific cleavage or other programmed activity. In genetics research, careful design helps researchers modify, regulate, or investigate gene function while reducing unintended genomic interactions; computational screening and experimental validation can assess candidate guides before use. Design choices therefore influence editing efficiency, specificity, and the interpretability of genetic experiments.

RNA Interference

2019

RNA interference (RNAi) is a process in which a small non-coding RNA molecule blocks the post-transcriptional expression of a gene by binding to its messenger RNA (mRNA) and preventing the protein from being translated. This process occurs naturally in cells, often through the activity of genomically-encoded microRNAs. Researchers can take advantage of this mechanism by introducing synthetic RNAs to deactivate specific genes for research or therapeutic purposes. For example, RNAi could be used...
