Antibody Effect Assay

An antibody effect assay is a laboratory method used to measure how antibodies alter the activity, survival, or infectivity of target cells or pathogens, providing functional evidence beyond antibody binding alone. In these assays, antibodies are incubated with a defined target, and their effects are quantified through mechanisms such as neutralization, inhibition of entry, opsonization, or antibody-dependent cellular cytotoxicity. Researchers use appropriate controls and dose comparisons to determine potency and activity under specific experimental conditions. Antibody effect assays support immunology and infection research by evaluating immune protection, comparing antibody candidates, characterizing pathogen responses, and guiding vaccine or therapeutic development.

A Multiplex Serological Assay for the Detection of Antibody Responses to Arboviruses

2025

Multiplex fluorescence immunoassays are becoming a reference for serology testing. Their application, however, is limited by the complexity and time-consuming bead-analyte coupling processes. This report presents a semi-automated, reproducible multiplex microsphere bead coupling protocol that is comparable to the standard method while considerably reducing time to results.

Functional Assessment of Antibody-Secreting B Cells Using ELISpot Assay

2025

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.

An Assay to Measure Influenza Neuraminidase Inhibition Antibody Titers

2025

This video demonstrates an assay measuring influenza neuraminidase (NA) inhibiting antibody titers in a serum sample. A serum sample containing antibodies against NA is added to a fetuin-coated microwell plate. Upon adding the virus, the antibodies prevent viral NA from cleaving fetuin and exposing its galactose moiety. A peroxidase-conjugated lectin is then added, which binds to the exposed galactose. The NA-inhibiting antibody titer is determined using a colorimetric test, where a peroxidase...

A Serum Bactericidal Assay for the Complement-Mediated Bactericidal Activity of Antibodies

2025

This video demonstrates a technique to evaluate the complement-mediated bactericidal activity of antibodies present in serum. This approach combines isolated serum with pathogenic bacteria and exogenous complement proteins. Upon incubation, the bacteria are grown on an agar plate, and the produced colonies are quantified to measure the serum bactericidal titer.

The Hemagglutination Inhibition Assay to Detect Serum Antibodies Against a Target Antigen

2025

The video demonstrates a hemagglutination inhibition assay for detecting serum antibodies against specific antigens. Confirmation of hemagglutination inhibition in the test well and hemagglutination in the control well is achieved through distinct visual patterns. This assay plays a crucial role in assessing immune response, vaccine effectiveness, and studying viral infections in virology and immunology.
