Cytokine Maturation

Cytokine maturation is the biological processing that converts inactive cytokine precursors into signaling molecules capable of coordinating immune and inflammatory responses. In many infection and immunology pathways, inflammasome activation recruits and activates caspase-1, which cleaves precursor forms such as pro-IL-1β and pro-IL-18 into their mature, secreted forms; other cytokines undergo distinct proteolytic or cellular processing steps. This regulation helps restrict potent inflammatory signals until danger-associated or pathogen-associated cues are detected. Understanding cytokine maturation clarifies how innate immune responses are initiated, amplified, and resolved, while supporting research into infection, autoinflammatory disease, vaccine responses, and therapeutic targeting of inflammatory pathways.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.
