Neutrophil Degranulation

Neutrophil degranulation is the regulated release of antimicrobial proteins and enzymes from intracellular granules, a central mechanism of innate immune defense against infection. Following recognition of microbial signals or inflammatory cues, receptor signaling activates the cytoskeleton and directs granules to fuse with the phagosome or plasma membrane, releasing substances such as proteases, antimicrobial peptides, and oxidant-generating components. These effectors help destroy engulfed pathogens and shape local inflammation, making degranulation important in host protection, infection research, and studies of inflammatory disease. Excessive or poorly controlled release can also injure surrounding tissues, highlighting its relevance to immune regulation and therapeutic development.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.
