O Antigen Staining

O antigen staining is an immunological method used to detect the O, or somatic, antigen on the surface of Gram-negative bacteria, where it forms the variable polysaccharide portion of lipopolysaccharide. The procedure uses antibodies that specifically bind O-antigen structures, with the resulting antigen-antibody complexes visualized through a fluorescent, enzymatic, or other detectable label. In immunology and infection research, this approach supports bacterial identification and serotyping, helps distinguish strains with different surface antigen profiles, and contributes to studies of host-pathogen interactions, transmission, and outbreak epidemiology.
