Phagocytosis Assay

A phagocytosis assay is a laboratory method used to measure how efficiently immune cells engulf and internalize particles, making it useful for evaluating innate immune function. In the assay, target particles such as microbes, beads, or labeled materials are incubated with phagocytes, and uptake is quantified through fluorescence, microscopy, flow cytometry, or related readouts. Researchers use these measurements to compare cell types, assess the effects of opsonization or infection, and determine whether pathogens alter engulfment and intracellular processing. This approach supports studies of host-pathogen interactions, immune deficiencies, antimicrobial responses, and potential immunomodulatory treatments.

This video demonstrates an assay to measure the antibody-dependent cellular phagocytosis (ADCP) by breast milk phagocytes. Fluorescently labeled microspheres conjugated to an antigen-antibody complex, are combined with a cell suspension isolated from breast milk to induce antibody-dependent cellular phagocytosis by the phagocytic leukocytes. Upon labeling the cells with antibodies against common leukocyte antigen (CD45), flow cytometry is used to measure the fluorescence intensity of...
