Phalloidin

Phalloidin is a bicyclic peptide that binds filamentous actin, making it a widely used probe for visualizing the cytoskeleton in fixed cells and tissues. It associates selectively with F-actin filaments and stabilizes them, while fluorescently labeled phalloidin enables their detection by fluorescence microscopy. In immunology and infection research, phalloidin staining reveals changes in cell shape, actin remodeling, migration, phagocytosis, and pathogen-associated cytoskeletal disruption. When combined with antibody-based markers, it helps distinguish host-cell structures from microbial components and relates actin organization to immune-cell function, cellular invasion, and disease mechanisms.

Source: Alluri, H., et al. Oxygen-Glucose Deprivation and Reoxygenation as an In Vitro Ischemia-Reperfusion Injury Model for Studying Blood-Brain Barrier Dysfunction. J. Vis. Exp. (2015).The video demonstrates the fluorescent localization of tight junction proteins and stress fibers in rat brain microvascular endothelial cells (RBMECs). In the stressed monolayer, fluorescently labeled antibodies are used to tag the tight junction proteins, while fluorophore-tagged phalloidin labels the stress...

Immunostaining to Visualize Viral Antigen and Host Protein Colocalization in the Insect Gut

Source: Zhang, L., et al. Immunofluorescent Labeling of Plant Virus and Insect Vector Proteins in Hemipteran Guts. J. Vis. Exp. (2021)The video demonstrates the preparation of gut tissue from a plant virus–infected insect vector for fluorescence imaging. Fixed guts were washed, permeabilized, and incubated with two fluorescent antibodies in a blocking solution—one targeting a viral antigen in vesicles, the other a vesicle-associated membrane protein. The tissue was counterstained for actin...

A Double-Labeling Immunofluorescence Technique to Visualize Host and Pathogen Proteins in an Infected Cell

2025

This video showcases double-labeling immunostaining with two different primary antibodies raised in the same species, binding specifically to parasite and host proteins. Furthermore, labeled secondary antibodies targeting these primary antibodies allow the distinct visualization of host and parasite proteins, facilitating the study of host-pathogen interaction.
