Tissue Supernatant

Tissue supernatant is the liquid fraction recovered from disrupted or homogenized tissue after insoluble cells, debris, and other particulate material are removed, making it useful for measuring soluble biological signals. During preparation, tissue is processed under defined conditions and centrifuged or otherwise clarified; the resulting fluid retains molecules released from or present within the tissue, including cytokines, antibodies, enzymes, and microbial products. In immunology and infection research, tissue supernatants support analyses of local inflammation, immune-cell activity, pathogen burden, and tissue damage, enabling comparisons between experimental conditions and linking molecular responses to disease progression or treatment effects.

An Assay To Evaluate the Anticancer Effects of Lactobacillus Cell-Free Supernatant

This video demonstrates an in vitro assay to study the anticancer effects of Lactobacillus cell-free supernatant (LCFS). Upon exposing colorectal cancer spheroids to varying doses of LCFS, the interaction with the bacterial metabolites in LCFS induces apoptosis in the cancer cells, disrupting the spheroid structure and leading to cell death in a dose-dependent manner.

Heparin Affinity Chromatography-Based Baculovirus Purification: A Technique to Isolate Baculovirus From Insect Cell Supernatant

In this video, we demonstrate the technique to isolate baculovirus from an insect cell supernatant using heparin affinity chromatography. The isolation is facilitated by the affinity between the heparin in the column to the baculovirus envelope glycoprotein.

Methods to create human 3D tumor tissues as test systems are described. These technologies are based on a decellularized Biological Vascularized Scaffold (BioVaSc), primary human cells and a tumor cell line, which can be cultured under static as well as under dynamic conditions in a flow bioreactor.

Here methods are presented to understand anti-cancer effects of Lactobacillus cell-free supernatant (LCFS). Colorectal cancer cell lines show cell deaths when treated with LCFS in 3D cultures. The process of generating spheroids can be optimized depending on the scaffold and the analysis methods presented are useful for evaluating the involved signaling pathways.
