Dmem 20% Fbs

DMEM with 20% FBS is a mammalian cell-culture medium made by supplementing Dulbecco’s Modified Eagle Medium with 20% fetal bovine serum, creating conditions that support cell maintenance and growth. DMEM supplies glucose, amino acids, vitamins, inorganic salts, and buffering components, while FBS contributes proteins, lipids, hormones, growth factors, and attachment-promoting factors that help cells survive and proliferate in vitro. In medicine and biomedical research, this formulation is used to expand cultured cells, maintain experimental models, and support studies of disease mechanisms, drug responses, and cellular behavior. Its defined serum concentration also helps standardize culture conditions across experiments.

This protocol describes a detailed workflow for the generation and ex vivo characterization of oncolytic viruses for expression of immunomodulators, using measles viruses encoding bispecific T cell engagers as an example. Application and adaptation to other vector platforms and transgenes will accelerate the development of novel immunovirotherapeutics for clinical translation.
