Ficoll Gradient Isolation

Ficoll gradient isolation is a density-based centrifugation technique used to separate peripheral blood mononuclear cells from whole blood for medical and biomedical research. Blood is layered over a Ficoll solution and centrifuged, causing components to migrate according to density: plasma remains above the gradient, mononuclear cells collect at the plasma-Ficoll interface, and granulocytes and erythrocytes move below it. Researchers then recover, wash, and resuspend the mononuclear cell layer for downstream analysis. This method supports immunophenotyping, flow cytometry, cell culture, infectious disease studies, transplant research, and investigations of immune function.

This protocol describes enzymatic digestion of mouse skin in nutrient-rich medium followed by gradient separation to isolate leukocytes. Cells thus derived can be used for diverse downstream applications. This is an effective, economical, and improved alternative to tissue dissociation machines and harsher trypsin and dispase-based tissue digestion protocols.
