Neonatal Lung Diseases

Neonatal lung diseases are conditions that impair breathing and gas exchange in newborns, particularly during the transition from fetal to independent respiration. They can arise when immature lungs produce insufficient surfactant, retain fetal lung fluid, or sustain injury from infection, inflammation, or disrupted development, leading to reduced alveolar stability and oxygen delivery. Clinical evaluation combines respiratory assessment with imaging and blood-gas measurements to identify the underlying problem and guide treatment. Management may include oxygen therapy, respiratory support, surfactant replacement, and infection control, while ongoing research aims to improve early diagnosis and reduce complications affecting long-term lung development.

This protocol describes the development of a neonatal piglet model of acute lung injury that models early pathogenic events occurring in the preterm lung, including sub-adequate surfactant quantity, hyperoxia, high-pressure ventilation, and inflammation, to facilitate understanding of molecular triggers of bronchopulmonary dysplasia and enhance therapeutic translation.

Lung ultrasound is a noninvasive and valuable tool for bedside evaluation of neonatal lung diseases. However, a relative lack of reference standards, protocols and guidelines may limit its application. Here, we aim to develop a standardized neonatal lung ultrasound diagnostic protocol to be used in clinical decision-making.

This technique of instilling drugs directly into the trachea of neonatal rodents is important in studying the impact of locally administered drugs or biologicals on neonatal lung diseases. Additionally, this method can also be used for inducing lung injury in animal models.
