Osteoarthritis Pain

Osteoarthritis pain is discomfort arising from a degenerative joint disorder that affects cartilage, bone, synovial tissues, and surrounding structures, often limiting movement and daily function. As cartilage becomes damaged, altered joint mechanics, inflammation, and sensitization of pain-sensing pathways can produce pain during activity as well as stiffness after rest; symptoms may persist even when structural changes are modest. Understanding these mechanisms supports clinical assessment and guides management through exercise, weight management, physical therapy, medications, and, when necessary, joint replacement. Studying osteoarthritis pain also informs research into targeted treatments that improve mobility and quality of life.

Osteoarthritis (OA), or degenerative joint disease, is a debilitating condition associated with pain that remains only partially controlled by available analgesics. Animal models are being developed to improve our understanding of OA-related pain mechanisms. Here we describe the methodology for the monoiodoacetate model of OA pain in the mouse.

We describe a protocol for the destabilization of the medial meniscus (DMM) model in mice, an effective tool for osteoarthritis (OA) research. In addition, we have demonstrated that deficiency of progranulincan exaggerate OA development and progression by using this model, indicating that progranulin plays a protective role in the pathogenesis of OA.

Diffuse noxious inhibitory control, temporal summation and wound hyperalgesia testing are demonstrated in the obstetric patient. These tests evaluate inhibitory and excitatory mechanisms of pain processing and are here utilized to evaluate endogenous analgesia at different time-points during pregnancy and the peripartum period to help reveal individual s risk for persistent pain.
