Stimulation Threshold

Stimulation threshold is the minimum intensity or duration of a stimulus required to produce a measurable physiological response, making it an important measure of tissue excitability in medicine. In excitable tissues, electrical stimulation changes the membrane potential; when depolarization reaches a critical level, voltage-gated ion channels activate and generate an action potential, with the threshold influenced by pulse duration, electrode placement, and tissue condition. Clinicians and researchers measure stimulation thresholds in nerve conduction studies, cardiac pacing, neuromodulation, rehabilitation, and sensory or motor testing. These measurements help identify altered excitability, calibrate therapeutic stimulation, and improve treatment safety and effectiveness.

Here, we describe a rapid reliable and simple procedure to determine the lowest temperature at which rats or mice show nocifensive behavior, i.e. the thermal nociceptive threshold (TNT). This method applies a slowly increasing thermal stimulus allowing precise and reproducible estimation of TNTs with minimum, if any, stress to the animals.

Psychophysical methods such as the QUEST estimation procedure can efficiently yield robust estimates of the stimulation intensity at which nonpainful sensations transition into painful sensations. By stimulating repeatedly at the threshold intensity, the variability in rating responses can directly be attributed to perceptual classifications in subsequent analyses.
