Tissue Necrosis Analysis

Tissue necrosis analysis is the systematic assessment of localized tissue death, helping clinicians and researchers determine the extent, characteristics, and likely consequences of injury. Necrotic cells lose membrane integrity, swell, and release intracellular contents, which can trigger surrounding inflammation; analysis identifies these changes through methods such as histopathology, microscopy, imaging, and selected biochemical markers. In medicine, the approach supports evaluation of ischemia, infection, trauma, tumors, and treatment-related injury. Accurate characterization can guide diagnosis, estimate disease severity, monitor treatment response, and distinguish irreversible damage from potentially viable tissue, improving clinical decision-making and advancing research into tissue repair and disease mechanisms.

Necrosis is considered as an “accidental” or unexpected form of cell death that ends in cell lysis. The first noticeable mention of “necrosis” was in 1859 when Rudolf Virchow used this term to describe advanced tissue breakdown in his compilation titled “Cell Pathology”. Morphological Manifestations of Necrosis Necrotic cells show different types of morphological appearance depending on the type of tissue and infection. In coagulative necrosis, cells become anucleated and die, but their...

Source: Chen, Y. et. al., Evaluating Virulence and Pathogenesis of Aeromonas Infection in a Caenorhabditis elegans Model. J. Vis. Exp. (2018)This video demonstrates a muscle necrosis assay in C. elegans (Caenorhabditis elegans) using pathogenic Aeromonas strains. By feeding green fluorescent protein (GFP)-expressing worms on Aeromonas-seeded plates, the assay models bacterial infection and reveals tissue damage through fluorescence imaging. The progressive loss of muscle structure and GFP...

Necrosis is a form of irreversible cell death caused by severe injury such as ischemia, toxins, or trauma. Unlike programmed cell death, it is an uncontrolled, pathological process that typically provokes inflammation in surrounding tissues.Pathophysiologic ChangesNecrosis begins when cells sustain critical damage, leading to swelling of organelles, particularly mitochondria, and rapid ATP depletion. As energy levels decline, membrane ion pumps fail, leading to calcium influx and eventually,...
